Experiment: TLR1 is labeled with Cy3 (a fluorophore) and TLR2 is labeled with Cy5 (a another fluorphore). Cy3 is excited by light of 540 nm and fluoresce, but Cy5 is not excited. Cy3 and Cy5 both show florescent resource energy transfer FRET.

Question: Based off this setup. If the researcher were to isolate macrophages carrying labeled TLRs and it shows no FRET response under light of 540 nm. These when they are exposed to peptidoglycan, a significant FRET response is observed along with an immune response signal. What do you think is occurring with the TLRs at the membrane to cause a immune signaling pathway to active in the macrophages.

Ans- peptidoglycan is such a polymer which is consisted of the amino acids and sugars. It is mainly found in the cell wall of some bacteria. So the treatment with the peptidoglycan initiate immune signaling as these treated cell will be taken as bacterial cell by the TLR. So the signal produced will be a mixture of FRET along with immune signaling.
