1.genetic transformation means acquiring a new gene in the cells of bacteria.In order to determine the cell is transformed we need to keep the cell in a particular enviroment.For example if we want to check transformation of antibiotic resistant gene then we need to place the cells in the medium containing the antibiotic.If the cells had transformed then they will grow on the medium containing antibiotic, if not then the cells will not survive.

So to compare, the cells grown on normal agar should be compared with the cells grown on antibiotic medium.

2.control plates are used for comparison of experimental results.A positive control will be showing intact plasmid carrying the desired selectable marker (e.g., antibiotic resistance) and negative control will be showing the cells only no DNA.Negative and positive controls should be included in the transformation step to evaluate the success of the experimental procedure.
