Question
Aunt Jenny, age 110, is planning to prepare her patented fruit cake for the holidays, but shemisplaced the family recipe and cannot remember what to add. Searching in the attic, she discovered the old
baking pan that has not been used for the last 30 years. It has still some of the baked-on crust of a fruit cake
stuck on the bottom. Aunt Jenny submits the crust to the local laboratory and has received a DNA sequence as
result of their investigation. This sequence is available to you as SEQUENCE-1.fasta on MOODLE. It is your
job to find out which fruits have been used in the old family recipe. Describe each step of your investigation.

Explanation / Answer
Copy the sequence. Go to NCBI BLAST: http://blast.ncbi.nlm.nih.gov/Blast.cgi?PROGRAM=blastn&BLAST_PROGRAMS=megaBlast&PAGE_TYPE=BlastSearch&SHOW_DEFAULTS=on&LINK_LOC=blasthome Make sure the "database: is set to "nucleotide sequence (nr/nt). Paste the entire sequence into the big empty box at the top of the page that says "Enter Query Sequence." Scroll to the bottom and hit the "BLAST" button on the bottom left. Now, wait, just as the message says, for the computer to give you results. Scroll past the image with all the red horizontal lines to the section with lines of text. The section is title "Descriptions." The first 2 words under the "Description" column of the "Descriptions" section are the scientific names of the fruits identified. Google the scientific name to get the common name. For example, the first fruit that should come up is: Actinidia deliciosa . A google search reveals that this is a kiwi.
