For the first part of the experiment, in which you are varying the amount of tyrosinase, how many milliliters of 15mM L-DOPA will be in each tube? 2. If 1 give you two tubes and tell you that one is enzyme and the other substrate, what experiment will you do to determine which is which? For this, you have no other source of enzyme or substrate to use, but you do have spec tubes and a spectrophotometer.
Explanation / Answer
we have to add 5 milliliters of 15 mM L-DOPA in each tube. the amount of L-DOPA is fixed and the amount of tyrosinase and phosphate buffer changes.for 5 reading tyrosinase amount = 0.1 ml,0.2 ml,0.3 ml,0.4 ml,& 0.5 ml. and the amount of phosphate buffer is= 2.9 ml, 2.8 ml, 2.7 ml, 2.6 ml, & 2.5 ml respetevely....and the final volume will be 3.0 ml plus 5mM OF 15mM L-DOPA...
