Question

I want to add a certain concentration of IPTG to 3 different reactions of a volume = 3.2mL. I have a stalk solution of 1M IPTG. I want to add 1mM IPTG to reaction 1, 0.5mM IPTG to reaction 2 and 0.25mM to reaction 3.
Outline how you would make a serial dilution of the IPTG, and how much of each dilution you would add to the reactions (hint: it's the same volume from each serial dilution). Explanation / Answer

Stock IPTG solution = 1 M

solution 1,

volume of stock needed = 0.001 M x 3.2 ml/1 M = 0.0032 ml = 3.2 microliter

volume water = 3200 - 3.2 = 3196.8 microliter = 3.1968 ml

solution 2,

volume of stock needed = 0.0005 M x 3.2 ml/1 M = 0.0016 ml = 1.6 microliter

volume water = 3200 - 3.2 = 3198.4 microliter = 3.1984 ml

solution 3,

volume of stock needed = 0.00025 M x 3.2 ml/1 M = 0.0008 ml = 0.8 microliter

volume water = 3200 - 3.2 = 3199.2 microliter = 3.1992 ml
