Question

Determination of the Molar Absorption Coefficient for Nucleosides MIROOTbeebso nd a nucleotide? 2. What is the Amax and the absorption coefficient (at the Amax) for each of the following nucleosides (Guanosine, Cytidine, Uridine, Adenosine, Thymidine and Inosine)? Make sure that you cite your literature source for this information. Absorption Coefficient Nucleoside Amax (nm) Reference 3. You have a stock solution of Inosine that when diluted with water 1:20 will give an absorbance at 248.5 nm of approximately 0.85. What was the concentration of the stock solution of Inosine in mol/L?

Explanation / Answer

1.)Nucleosides are glycosylamines consisting of a nucleobase bound to ribose or deoxyribonucleic ribose sugar via a beta-glycosydic linkage. Cytidine , thymidine , guanosine are some examples .

Nucleotide is composed of nucleobase , a nitrogenous Base of five carbon sugars and one two or three phosphate groups forming bonds with 2,3 or 5 th carbon of sugar . 5 carbon site is most common .

Nucleic acid bases are nitrogen containing biological compounds that form nucleoside which in turn are compounds of nucleotide together are monomers constituting basic building block of nucleic acids . Purine and pyramidine bases together forms it .

3.)By beer lambert law we know that

A=£cl , where £ is molar extinction coefficient , A is absorbance , l is pathlength and c is concentration .

Molar extinction coefficient and pathlength is needed to calculate concentration . Here 248.5nm is just indicating that there is a possible absorption .

By referring data from --Urban M, joubert N, purse n etal mechanism by which human DNA chooses to polymerase a nucleoside triphosphate (2010)biochem (49):727-735-- data path length taken as 1 cm and £=12300Lmol-1cm-1

C=0.85/1*12300*0.01

=0.85*0.01 mol/litre .
