Phosphate is readily reacted with ammonium molybdate in the presence of some suitable reducing agents and gives a blue colored complex.

Phosphate + ammonium molybdate ===) phosphomolybdates

Phosphomolybdates + Ferrous ammonium sulfate ===) molybdenum blue.

The intensity of blue colored solution is increased with the concentration of the phosphate content in the sample. The blue colored complex absorbance is measured by colorimetric technique. The unknown sample phosphate content value is measured by calibration curve of a series of known concentration of phosphate in the complex form of molybdenum blue.
