Question

A recent review article outlined key features of peptides capable of forming a stable beta-sheets. For each of the features listed below, explain why each feature is likely stabilizing to these motifs.

a) The peptide should contain the following sequence: Asn-Pro-Asp-Gly

b) There should be favorable cross-strand side chain–side chain interactions between residues on the same face of the sheet. The most favorable include TrpTrp interactions and Lys-Glu interactions. (Include a drawing of these two specific interactions in your answer.

c) There should be a disulfide bond, distal to the beta-turn.

Explanation / Answer

a) The peptide should contain the following sequence: Asn-Pro-Asp-Gly because proline a secondary amino acid and causes distortion in the secondary structure due to the distorted geometry and aspartate has apolar group which also cause a steric hinderance and leads to beta turn.

b) The most favorable include Trp-Trp interactions and Lys-Glu interactions because the first one leads to a hydrogen bond intractions and the letter one leads to formation of a salt briegde. This causes a stabilization in the sheets.

c) There should be a disulfide bond, distal to the beta-turn because this causes a distortion in the peptide bond

plane and a turn is likely to take place.
