If instead of assaying for resistance to rifampicin we had assayed for conversion from histine auxotrophy to prototrophy (as is done in the Ames test) how would our mutation frequencies have differed? Why?My results from my lab were 500 cfu for the Rif cells incubated in light conditions after the uv exposure and 112.5 cfu for the Rif cells incubated in dark conditions after th uv exposure.
Please explain why and how the mutation frequencies would change and what would happen theoretically to the repair frequency in a AMES test.

Explanation / Answer
In the AMES test histine auxotrophy is caused by point mutations in strain TA-1531 while rifampicin resistance is due to changes in one or multiple clusters among 3 clusters in the RNA polymerase where rifampicin binds. Hence the probability of reversing Rifampicin resistance is much higher than histine auxotrophy.
