Explanation / Answer

TLC is a qualitative technique to identify the number of compounds in a given sample compound.

1. If given sample is more concentrated, after its run in solvent it is getting blurred, so that we can not identify the presence/ or absence of compounds data.

2. If we spot too low compound on TLC plate, we can not identify low concentrated compound in the given sample.

3. Ethylene glycol is very polar one like methanol. If compound does not move up even in ethylene glycol, it means compound is very polar and it bins on silica plate.

4. You need to use water, methanol system an have to check at various proportions of both. Because those compounds do not lift up in such polar solvents (mthanol, ethylene glycol) bind firmly to the plate. So you need to use water fractions of these solvens. Moreover, based on nature of solvents, we can change solvent system. For acidic compounds you need to add a pinch triethyl amine to the mobile phase.

5. TLC is the primary technique to know the nature of compounds in unknown sample.

the compounds at the top of the TLC comes fast from the coloumn with low polar mobile phase. Those compoun at the down of TLC plate needs more polar mobile phase to elute in coloumn chromatography.
